- Epitope tagging (e.g. His-tag) is a technique in which a known epitope is fused to a recombinant protein using genetic engineering. By choosing a particular epitope and recombinant protein combination, epitope tagging makes it possible to detect proteins for which no antibody is available. The 6x His tag is a synthetic oligo peptide consisting of 6 consecutive histidine residues (HHHHHH). A variety of plasmids contain DNA that encodes an amino- or carboxy-terminal tag consisting of six histidine (6xHis) residues followed by an extended multiple cloning site. These plasmids facilitate expression of His-tagged recombinant proteins that can be isolated or purified by immobilized metal affinity chromatography. Applications: Suitable for use in Western Blot, Immunohistochemistry, Flow Cytometry, Immunoprecipitation and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:5000 Immunohistochemistry: 1:100 Immunocytochemistry: 1:100 Immunoprecipitation: 1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
