- N-methyl-D-aspartate receptor subunit NR1 (NMDAR1) is a multi-pass membrane protein belonging to the glutamate-gated ion channel family and is one of two subunits comprising the tetrameric protein NMDAR localized throughout the brain at excitatory synapses. NMDAR1 is mediated by glycine and is a critical component to excitotoxicity, learning, memory acquisition, synaptic plasticity, and synaptogenesis. It also plays a regulatory role for neuronal functions involving glutamate neurotransmission and the targeting of NMDA receptors on the cell surface. As a glutamate-gated ion channel, NMDAR1 is highly calcium permeable and possesses sensitivity to magnesium in a voltage-dependent manner. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 0.1ug/ml detected NMDAR1 on 10ug of mouse microsomal brain tissue lysate. Immunohistochemistry (paraffin): 1:250 detected NMDAR1 in rat cerebellum tissue. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
