Extra Details:
Luciferase is a generic term for the class of oxidative enzymes used in bioluminescence and is distinct from a photoprotein. Luciferase catalyzes a bioluminescent reaction which requires the substrate luciferin as well as Mg2+ and ATP, produces green light with a wavelength of 562nm. Luciferase from firefly is widely used as a reporter for studying gene regulation and function, and for pharmaceutical screening. Recombinant protein corresponding to firefly Luciferase, expressed in E. coli. Molecular Weight: ~100kD, a dimer of two subunits of ~50kD Specific Activity: 15-30x10e6 light units/mg protein Unit Definition: One light unit produces a biometer peak height equivalent to 0.02 microcuries of 14C in PPO/POPOP cocktail. Light units are measured in 50 microliters of assay mixture containing 5 picomoles of ATP and 7.5 nanomoles luciferin in glycine-Tris buffer, pH 7.6 at 25°C. Light units are measured in a 50ul assay mixture containing 5 pmole of ATP and 7.5 nmole of luciferin in Tris-glycine buffer at pH 7.6 at 25°C. Enzymatic Impurities: ATPase: <5nm/mg Nucleoside Diphosphokinase: <20nm/mg Sensitivity Test: < or = 1 femtomole of ATP can be determined using 0.2micrograms of luciferase and suitably sensitive liquid scinitillation counters or luminometers. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
