- Extra Details:
- Luciferin 4-monooxygenase (UniProt: P08659; also known as EC: 1.13.12.7, Luciferase) is a peroxisomal enzyme that belongs to the ATP-dependent AMP-binding enzyme family. It catalyzes the production of oxyluciferin from luciferin in an ATP and magnesium dependent reaction and releases a photon of light when oxyluciferin returns to the ground state. This reaction proceeds in two steps where the first step involves adenylation of luciferin and in the second step luciferase acts as an oxygenase on adenyl-luciferin to produce oxyluciferin and carbon dioxide, the decay of oxyluciferin producing a photon of light of 562 nm. Luciferase from the firefly is commonly used in reporter assay where the total light emission is proportional to the luciferase activity of the sample. Applications: Suitable for use in Immunocytochemistry and Western Blot. Other applications not tested. Recommended Dilution: Immunocytochemistry: 1:500 detected Luciferase in HeLa cells transfected with Luciferase expression plasmid. Western Blot: 1:1000 detected Luciferase in HeLa cells transfected with luciferase expression plasmid. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
