- NR2F6 (Nuclear receptor subfamily 2 group F member 6, EAR-2, COUP-TFIII) is a member of the NR2F family of transcription factors, defined by their role in the regulation of organogenesis, neurogenesis, and cellular differentiation during embryonic development (1,3). NR2F6 acts as a negative regulator for TRβ1-mediated transcriptional activity, a transcriptional repressor of IL-17 expression in Th-17 differentiated CD4+ T cells, and a repressor of the NFAT/AP-1 complex on both the interleukin-2 and the interferon-gamma cytokine promoters (2-5). NR2F6 has a non-redundant role in antagonizing anti-cancer effector T cell responses (5). It has been shown that loss of NR2F6 enhances the activity of established PD-L1 checkpoint blockade, promoting tumor regression and increased survival in mouse models (5,6). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% nonfat dry milk, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
