- Tumor necrosis factor (TNF, cachexin or cachectin and formerly known as tumor necrosis factor-alpha) is a cytokine involved in systemic inflammation and is a member of a group of cytokines that stimulate the acute phase reaction. This is a high activity construct in which two trimeric TNF-alpha ligands are artificially linked via the collagen domain of ACRP30. Sequence: The extracellular domain of human TNF-α (aa85-233) is fused at the N-terminus to mouse ACRP30headless (aa18-111) and a FLAG®-tag, expressed in HEK 293 cells. Molecular Weight: ~34kD (SDS-PAGE) Biological Activity: Activates human and mouse TNF-R1 and TNF-R2. Induces cell death at a concentration range of 0.01-10ng/ml (ED50= 0.05ng/ml or 2x10e7 Units/mg; WEHI 164 cells). Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
