- The ST2 gene was originally identified as a gene induced by serum or oncogene expression in fibroblasts. The gene produces a shorter soluble secreted form (ST2) and a longer, transmembrane form (ST2L) by alternative splicing. Soluble ST2 has been shown to downregulate the expression of TLR1 and TLR4. ST2L negatively regulates TLR4 signaling and induces endotoxin tolerance, and enhances Th2 responses. IL-33 is the specific ligand for ST2L. Applications: Suitable for use in Immunohistochemistry, ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA (direct or indirect): 1:2000-1:5000 Western Blot: 1:2000-1:5000 using ECL. Suggested blocking and dilution buffer is PBST with 0.05% Tween 20 and 5% skim milk. Suggested incubation time is 1 hour at room temperature Immunohistochemistry (paraffin sections): 1:50-1:200 Optimal dilutions to be determined by researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
