- IgG2 is mainly used to neutralize antigens or block receptor ligand binding, and its CDC and ADCC effects are very weak, and only EGFR antibodies were marketed based on IGG2 in the early days. However, with the rise of immune checkpoint research, more and more IGG2-based drugs have entered the clinic and entered the market. Although IgG2 has weak binding with C1q, it can still trigger CDC effect when the antigen or antibody solubility is high, and IgG2 is the only subtype that can bind FC-G Gammariia (CD32a). Moreover, this binding can be regulated by single nucleotide polymorphisms (SNPS), which can affect the functional activity of IgG2.
