- Extra Details:
- Interleukin-5 (IL-5) is a homodimeric cytokine, where two identical 126-amino-acid monomers are linked by a disulfide bond. It is primarily secreted by activated Th2 cells, with additional sources including eosinophils, mast cells, iNKT cells, and NK cells.IL-5 is the principal regulator of eosinophils, driving their differentiation, survival, activation, and chemotaxis. It also activates basophils. Beyond granulocytes, IL-5 acts on B cells to promote immunoglobulin production, growth, and differentiation, linking it to humoral immunity. Its biological effects are mediated by a cell surface receptor complex composed of a unique IL-5RA Alpha chain and the common beta chain (B Betac/CD131). Receptor engagement activates JAK2 and other kinases, triggering downstream JAK-STAT5 and RAS-MAPK signaling pathways. Bioavailability is naturally regulated by soluble IL-5RA Alpha, which acts as a decoy receptor. Thus, IL-5 is a central cytokine in type 2 immunity, allergic inflammation, and antibody responses, making it a key therapeutic target for eosinophil-driven diseases.
