Plasminogen is a glycoprotein present in human plasma at approximately 12 mg per 100 ml and serves as the precursor to plasmin, a key serine protease in fibrinolysis. Synthesized in the liver, plasminogen circulates in a closed, activation-resistant conformation until it binds to fibrin or cell surfaces, where it is converted to active plasmin by tissue plasminogen activator (tPA) or urokinase. Plasminogen contains multiple kringle domains that facilitate its binding to lysine residues on fibrin and cell surfaces, enhancing its localization and activation. Functionally, plasminogen is essential not only for the dissolution of blood clots but also for tissue remodeling, inflammation, cell migration, and the removal of misfolded or aggregated proteins. Deficiency in plasminogen, particularly type I (hypoplasminogenemia), leads to rare disorders such as ligneous conjunctivitis, characterized by pseudomembrane formation on mucosal surfaces and impaired wound healing. Therapeutically, plasminogen supplementation is used to treat plasminogen deficiency, and its activation pathway is targeted by thrombolytic drugs for conditions like stroke and myocardial infarction.
