- Smads are a family of intracellular proteins that transmit transforming growth factor beta (TGF-b) superfamily signals from the cell surface to the nucleus. The Smad family is divided into three subclasses: receptor regulated Smads, (Smads 1, 2, 3, 5 and 8); the common partner, (Smad4) that functions via its interaction to the various Smads; and the inhibitory Smads, (Smads 6 and 7). SMAD7, also known as Mothers Against Decapentaplegic homolog 7 (MADH7), inhibits selected pathways by binding directly to cell-surface receptors and preventing the activation-induced phosphorylation of other Smad subunits. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml tested with lysates of H9 human cutaneous T lymphoma cell line, PT18 mouse mast/basophil cell line, and Rat-2 rat embryonic fibroblast cell line Immunohistochemistry: 8-25ug/ml in immersion fixed paraffin-embedded sections of human liver array or liver cancer tissue Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
