Weitere Details:: SGK1 is a member of the serum- and glucocorticoid-induced protein kinase family. SGK1 is activated in vitro by 3-phosphoinositide- dependent protein kinase-1 (PDK1) and in vivo in response to signals that activate phosphatidylinositol (PI) 3-kinase. SGK1, mRNA is expressed in all tissues and the level of SGK1 mRNA is increased by stimulation with serum or dexamethasone. Applications: Suitable for use in Western Blot. Other applications have not been tested. Recommended Dilutions: Western Blot: 1:1000-1:3000. Not suitable for use with cell lysates. Detects phospho- SGK1 after phosphorylation of unactive SGK1 (S1010-82) with PDK1. Unactive SGK1 was incubated with or without PDK1 for 30 min at 30ºC. Phosphorylation was termiated by boiling in reducing sample buffer. Aliquots (50ng) were resolved by electrophoresis, transferred to nitrocellulose and probed with S1010-84 (1:3000). Proteins were visualized using an IgG (HRP) goat anti rabbit secondary antibody and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
