- This gene encodes a DNA-binding protein that has high specificity for scaffold or matrix attachment region DNA elements (S/MAR DNA). This protein is thought to be involved in attaching the base of chromatin loops to the nuclear matrix but there is conflicting evidence as to whether this protein is a component of chromatin or a nuclear matrix protein. Scaffold attachment factors are a specific subset of nuclear matrix proteins (NMP) that specifically bind to S/MAR. This encoded protein is thought to serve as a molecular base to assemble a 'transcriptosome complex' in the vicinity of actively transcribed genes. It is involved in the regulation of the heat shock protein 27 transcription and also can act as an estrogen receptor corepressor. This gene is a candidate gene for breast tumorigenesis. Applications: Suitable for use in Immunohistochemistry, Immunocytochemistry, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Reported to stain SAF-B in paraffin tissue sections. Immunocytochemistry: Reported to stain SAF-B in MCF-7 cells. Immunoprecipitation: Reported to immunoprecipitate SAF-B from MCF-7 and HepG2 cell lysates. Western Blot: 0.5ug/ml detects SAF-B in whole cell extracts or RIPA lysates from MCF-7 cells. MCF-7 whole cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S0430 (0.5ug/ml). Proteins were visualized using an IgG (HRP) goat anti-mouse and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
