- Protein tyrosine phosphatase 1B (PTP1B), also known as protein tyrosine phosphatase, non-receptor type 1 (PTPN1) and placental protein tyrosine phosphatase, dephosphorylates tyrosine residues in proteins. PTP1B is a cytosolic enzyme anchored in the endoplasmic reticulum that plays a critical role in regulating growth factor responsiveness. PTP1B knockout mice show increased resistance to high fat diet-induced obesity and have exaggerated responses to insulin. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Knockout Validated: PTP1B is specifically detected in HeLa human cervical epithelial carcinoma parental cell line but is not detectable in PTP1B knockout HeLa cell line. Western Blot: 0.05-0.3ug/ml using TF‑1 human erythroleukemic cell line and CHP‑100 human neuroblastoma cell line Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
