- Activation of PKC is one of the earliest events in the cascade leading to a variety of cellular responses such as secretion, gene expression, proliferation and muscle contraction (1,2). Protein kinase D (PKD), also called PKCmu, is a serine/threonine kinase (3,4). Activation of PKD/PKCmu is dependent on the phosphorylation of two activation loop sites, namely serine 744 and serine 748, via a PKC-dependent signal transduction pathway (5). In addition to the two activation loop sites, the C-terminal serine 916 has been identified as an autophosphorylation site for PKD/ PKCmu. Phosphorylation at the Ser916 site correlates with PKD/PKCmu catalytic activity (6). Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Immunoprecipitation: 1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
