- PKCa (Protein kinase C alpha) is an 80kD member of the PKC subfamily, AGC Ser/Thr protein kinase family of enzymes. It is widely expressed, and serves multiple cell-specific functions. PKCa is activated by increased diacylglycerol and intracellular calcium, and translocates to several sites such as the Golgi, nucleus and plasma membrane. Human PKCa contains two zinc finger regions (aa36-151), a protein kinase domain (aa339-557), and an AGC kinase region (aa598-668). Phosphorylations on Thr497, Thr638 and Ser657 are necessary for kinase activity. over aa604-672, human PKCa shows 100% aa identity to mouse PKCa. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml in human 293T, Saos-2, mouse Balb/c-3T3 cells Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
