- Myelin protein zero (P0) is the most abundant protein in mammalian peripheral nerve myelin. An interaction between P0 and PMP22 has been detected in compact myelin. Defects in PMP22 have been related to Charcot-Marie-Tooth disease (CMT) and inflammatory demyelinating neuropathy (IDP). Immunization with PMP22 induces experimental auto-immune neuritis without central nervous inflammation. Therefore, PMP22 is considered as candidate auto-antigen in inflammatory diseases of the peripheral nervous system, including both acute and chronic inflammatory demyelinating polyradiculoneuropathy. Applications: Suitable for use in Western Blot, Immunohistochemistry and ELISA. Other aplications not tested. Recommended Dilutions: Western Blot: 1:50 Immunohistochemistry (Paraffin): 1:50 Hydrogen peroxide treatment to reduce endogenous peroxidase acctivity. Requires HIER using Tris-EDTA buffer pH6.5 prior to staining. Blocking was done in 10% goat serum in TBS. Antibodies were incubated in TBS, 1% BSA at RT for 1hr. Optimal dilutions to be determined by researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
