- Paxillin is a 65kD cytoskeletal adaptor protein and member of the Paxillin family. Human Paxillin is 591 aa in length and contains four LIM zinc-binding domains. Alternative splicing produces three isoforms. Human Paxillin is 94% and 85% identical to mouse and rat Paxillin, respectively. Paxillin is found at the interface between actin filaments and the plasma membrane, and it localizes to focal adhesions, where it provides a platform for the integration and coordination of adhesion- and growth factor-related signals. Paxillin phosphorylation at tyrosines 31 and 118 is required for integrin-mediated cytoskeletal reorganization, and may play a role in the disassembly of focal adhesions and stress fibers during cellular transformation. Increased Paxillin protein levels have also been reported in several malignancies, including renal, lung, and breast carcinomas. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 2ug/ml with K562 human chronic myelogenous leukemia cell line and Jurkat human acute T cell leukemia cell line treated with Pervanadate. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
