- Nucleolin is an abundant, 106kD nucleolar phosphoprotein. It is the major protein in actively dividing cells, whereas the degraded forms are relatively abundant in non-dividing cells. Stability of the nucleolin protein is cell proliferation-dependent. It plays a role in rDNA transcription, organization, and rRNA processing. It may alter DNA organization in response to cell cycle controls. It is also a good mammalian nucleolus marker. Applications: Suitable for use in ELISA, Immunoprecipitation, Immunocytochemistry and Western Blot. Other applications not tested. Recommended Dilutions: Immunoprecipitation: Reported to immunoprecipitate nucleolin from a MDCK total cell lysate. Immunocytochemistry: 1:10-1:100 showed positive immunostaining for nuceolin in HeLa cells fixed with 4% paraformaldehyde and permeabilized with 0.25% Triton-X 100. Western Blot: 1:1000-1:5000 HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with N6945 (1:5000). Proteins were visualized using an IgG (HRP) goat anti-mouse and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
