- Human Lymphotoxin beta receptor (LTbR, TNFSF3) is a member of the TNF receptor superfamily with similarilty to CD120a(TNFR1) and CD120b(TNFR2). It is a expressed mainly on non lymphoid tissues and appears to have an important role in secondary lymphoid organ development. Suppression of LTbR signaling can alleviate autoimmunity or exasperate mycobacterial infection(6). Its ligands include Ltab2 and LIGHT. In mice, LTbR signaling is important for development and function of HEV, Dendritic cells(3), and Mast cells. Applications: Suitable for use in Flow Cytometry and ELISA. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50 (5ug/ml) Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 cultured human U-937 cells were pre incubated 5 min with 20ul of 250ug/ml human Ig (to block non specific binding). After which they were incubated 45 minutes on ice with 80ul of L8015-03G at a dilution factor of 1:50 (5ug/ml). 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 0.6 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. 4. Binding was blocked when cells were pre incubated with 20ul of 0.5mg/ml anti-LTbR antibody (L8015-03D). Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
