Weitere Details:
Integrins are heterodimeric proteins made up of alpha and beta subunits. At least 18 alpha and 8 beta subunits have been described in mammals. Integrin family members are membrane receptors involved in cell adhesion and recognition in a variety of processes including embryogenesis, hemostasis, tissue repair, immune response and metastatic diffusion of tumor cells. This gene encodes a beta subunit. Multiple alternatively spliced transcript variants which encode different protein isoforms have been found for this gene. Applications: Suitable for use in Flow Cytometry, Immunocytochemistry, Immunoprecipitatin, and Western Blot. Also suitable for inhibition of Jurkat binding to collagen and fibrinogen. Other applications have not been tested. Recommended Dilution: Flow Cytometry: 1:200-1:500 using live cells and goat anti-mouse secondary antibody. Immunocytochemistry: can be used with unfixed cells in culture. Oftentimes, fixing with 2-3% PFA for 5 minutes after primary antibody incubation will assist in obtaining strong signals since it fixes the primary antibodies to the tissues. Immunoprecipitation: 1:250-1:500. Western Blot: 1:1000-1:2000. (reducing, 115kD; non-reducing, 130kD) Inhibition of Jurkat binding to collagen and fibronectin: 1:100. Note: epitope is trypsin sensitive; cells should be prepared with PBS + EDTA (2mM) or other non-enzymatic methods. Blocking will not occur with trypsinized cells Note: Binding of antibody may be inhibited by magnesium ion concentrations greater than 0.5mM. Optimal dilution to be determined by the researcher. Control: Human lung, liver, and skeletal muscle tissues Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
