- H2AX is involved in DNA repair and the maintenance of genomic stability. although its precise role is still to be defined. H2AX is phosphorylated soon after the appearance of double-stranded breaks. and has been implicated both in homologous recombination and nonhomologous end joining DNA repair pathways. H2AX is thought to have a critical function in the recruitment of DNA repair factors and DNA damage-signaling proteins. Hyperphoshorylation of H2AX may be linked to chromatin fragmentation prior to apoptosis. Applications: Suitable for use in Immunohistochemistry, Western Blot and Peptide Inhibition analysis. Other applications not tested. Recommended Dilutions: Western Blot: 1:2000-1:10,000 detected histone H2A.X in acid-extracted proteins from HeLa cells. The antibody did not detect recombinant Histone H2A. Immunohistochemistry (paraffin): 1:1000 detected Histone H2A.X in human pancreas and human colon tissue sections. Peptide Inhibition Analysis: 1 and 10uM of the immunizing histone H2A.X peptide abolished detection of histone H2A.X by this lot in immunoblot analysis of HeLa acid extracts. Optimal dilutions to be determined by the researcher. Positive Control: HeLa acid extracts Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
