- Histamine [2-4-Imidazoyl)ethylamine] is produced by decarboxylation of its precursor, histidine, by histidine decarboxylase. It is widely distributed compound in peripheral tissues, where it influences as variety of physiological activities including inflammation, gastric acid secretion, and smooth muscle contraction. Histamine is localized in mast cells, neuronal cells, and neuroendocrine cells of the gastric tract, lung, and kidney, in cerebrovascular endothelial cell, and in the peripheral nervous system. It has potent vasodilatory and plasma extravasation effects on capillaries when secreted from mast cells in allergic hypersensitivity responses or inflammation. Histamine interacts with at least 3 receptor subtypes: H1, H2, H3 receptors that differ in their pharmacological properties. Applications: Suitable for use in ELISA. Other applications not tested. Recommended Dilutions: ELISA: 1:5000-1:20,000 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
