- Glycogen Synthase Kinase-3 (GSK-3) is a serine/threonine kinase initially identified as an inhibitor of glycogen synthase. Two isoforms (GSK-3α and GSK-3β) share 85% amino acid identity. GSK-3β, inhibited by phosphorylation at S9 by Akt, is involved in energy metabolism, body pattern formation, and neuronal cell development. Hybridoma: Produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a rat immunized with purified, E. coli-derived recombinant human Glycogen Synthase Kinase-3 beta (rhGSK3b; aa 1-420; Accession # P49841). Recommended Dilution: Western Blot: An antibody concentration of 1.0ug/ml is recommended. Optimal dilutions should be determined by the individual laboratory. Optimal dilutions to be determined by the researcher. Protocols for Immunoblotting: Blotting Buffer Blocking Solution Antibody Solution 25 mM Tris, pH 7.4 0.15 M NaCl 0.1% Tween® 20 5% nonfat dry milk in Blotting Buffer Adjust pH to 7.4 5% nonfat dry milk in Blotting Buffer Adjust pH to 7.4 Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
