Weitere Details:: FOXP3 (a 431 amino acid protein) is a member of the forkhead/winged-helix family of transcriptional regulators and is highly conserved across mammals. FOXP3 is essential for normal immune homeostasis. FOXP3 is stably and constitutively expressed at a high level in CD25 + CD4 positive regulatory T cells, at low level in CD4 positive/CD25 negative cells, and is absent in CD4 negative/CD8 positive T cells. FOXP3 may be a master regulatory gene and a more specific marker of regulatory T cells than other T cells. Applications: Suitable for use in ELISA, Western Blot, Flow Cytometry, Immunofluorescence/Immunocytochemistry and Immunohistochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:8000 Western Blot: 1-3ug/ml ~47kD band observed in human muscle lysates and in MOLT4 cell line lysates Immunohistochemistry (Parrafin): 0.3-1ug/ml Requires HIER using Tris-EDTA buffer pH 9.0 (Human tonsil, stains nuclei of some cells in the interfollicular area) Flow Cytometry: 10ug/ml Jurkat cells Immunofluorescence (IC): 10ug/ml U2OS cells Immunofluorescence (IC): 2-4ug/ml overnight human blood cells Optimal dilution to be determined by researcher. Negative Conrol (WB): Human pancreas lysates Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
