- Endothelial Cell Differentiation Gene-2, or EDG-2, encodes a 50kD protein belonging to the family of G-Protein Coupled Receptors (GPCR’s), and shows a strong affinity for LPA. To date, 8 members of the EDG receptor family have been identified. These receptors are suggested to have a role in modulation of growth-related effects including cell proliferation and differentiation as well as protective, anti-apoptotic effects. EDG receptor expression is developmentally regulated and the expression of the 8 members of this family of proteins appears to be tissue specific. EDG receptors couple to G-proteins to signal through Ras and MAPK pathways and other protein tyrosine kinase pathways. EDG-2 is also known as LPA1. The ligand for EDG-2 is lysophosphatidic acid (LPA). EDG-2 is expressed by oligodendrocytes and some human malignant T-cell lines. EDG-2 was originally identified in the ventricular zone of the developing cerebral cortex. Together with EDG-4, EDG-2 may have anti-apoptotic effects in cardiomyocytes subjected to hypoxia and adrenergic stimulation. LPA1 (also known as EDG-2) is one of four known LPA receptors (LPA1-4).1 LPA1-3 are part a family of G protein-coupled receptors that share identity to the sphingosine-1-phosphate receptors (S1P1-5). LPA1 couples with three types of G proteins, Gi/o, Gq, and G12/13 to induce a range of cellular responses including activation of phospholipase C, multiple kinases, the serum response element and cell proliferation. LPA1 mRNA is detected in a wide range of tissues and cells including brain, heart, small intestine, spleen, testis, kidney, HeLa cells, and A549 cells. Murine and human LPA1 have 364 amino acids with an estimated molecular weight of 41kD.
