Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation, Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5–2ug/ml detects CFTR in RIPA lysates from CFTR-transfected BHK cells. Reported to detect CFTR from a human T84 colon carcinoma epithelial cell lysate. CFTR-transfected BHK cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C9040-01 (0.5ug/ml). Proteins were visualized using IgG (HRP) goat anti-mouse secondary antibody and a chemiluminescence detection system. Note: Do not boil the lysate. Instead incubate at 37°C for 30 minutes. Immunoprecipitation: Immunoprecipitates CFTR from a T84 cell lysate. Immunocytochemistry: Reported to immunostain CFTR in T84 cells. Note: CFTR runs as a diffuse protein on SDS-PAGE. Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
