- Human Syndecan-1 (also known as CD138) is a variably glycosylated, dimeric, type I transmembrane (TM) protein that belongs to the Syndecan family. It is synthesized as a 310 amino acid (aa) precursor that contains a 17aa signal sequence, a 234aa extracellular domain (ECD), a 25aa TM segment, and a 34aa cytoplasmic region. The ECD shows various degrees of heparan sulfate and chondroitin sulfate modification, leading to native molecular weights for Syndecan-1 of 120-200 kDa. Proteolytic cleavage of the membrane-bound ECD yields soluble forms of approximately the same molecular weight. Syndecan-1 is an epithelial cell Syndecan involved in Wnt and chemokine signaling. Human Syndecan-1 ECD shares 71% aa identity with the ECD of both rat and mouse Syndecan-1. Applications: Suitable for use in Direct ELISA, Immunohistochemistry, Immunocytochemistry, DualRNAscope ISH-IHC Compatible and Flow Cytometry. Other applications not tested. Recommended Dilutions: Immunohistochemistry (Paraffin): 8-25ug/ml stains immersion fixed paraffin-embedded sections of human intestine and immersion fixed paraffin-embedded sections of human cervical cancer tissue. Flow Cytometry: 0.25ug/10e6 cells RPMI 8226 human mutiple myeloma cell line Immunocytochemistry: 3-25ug/ml Immersion fixed U266 human myeloma cell line. Dual RNAscope ISH-IHC Compatible: Immersion fixed paraffin-embedded sections of human colon. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing.
