Weitere Details:
Human CD137 Ligand is a type II transmembrane protein constitutively expressed by monocytes, B cells neuroblastoma cells (1), and some activated T cells. The interaction of CD137 (4-1BB) and CD137L can supply a costimulatory signal to T cells through CD137. This costimulus seems to be more effective for CD8+ effector T cells than for CD4+ helper T cells (3). The interaction can also effect a reverse signal, inducing monocyte activation (2), or T cell inactivation and programmed death (4) through CD137L. Applications: Suitable for use in ELISA, Western Blot and Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5ug/ml Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 cultured Raji cells were incubated 45 minutes on ice with 80ul C2548-55B at 5ug/ml. 2. Cells were washed twice and incubated with 2° reagent Streptavidin/R-Phycoerythrin (S973-88D). 3. Cells were washed three times, fixed and analyzed by FACS. 4. Cells stained positive with a mean shift of 0.79 log10 fluorescent units when compared to a Mouse IgG2a/Biotin negative control at a similar concentration. 5. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml C2548-55A. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
