- Human CD100 is a transmembrane glycoprotein that belongs to the semaphorin gene family and is found on most hematopoietic cells except RBC, platelets and immature bone marrow cells. CD100 plays a key role in lymphocyte activation. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1ug/ml Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 cultred human HPB-MLT cells were washed and preincubated for 45 minutes on ice with 80ul of C2445-25B at 1ug/ml. 2. Wash cells twice and incubate with Streptavidin (PE). 3. Wash cells three times, fix and analyze using FACS. 3. Cells were stained positive with a mean shift of 0.83 log10 flourescent units when compared to a mouse IgGM/Biotin negative control at a similar concentration. 4. Binding was blocked when pre-incubatde 10 minutes with 20ul of 0.5mg/ml CD100 (C2445-25A). Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
