- MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. CD94 cell surface antigen, also known as the Kp43 antigen, is expressed by NK cells and a subset of T lymphocytes. CD94 has been shown to exist in two forms, of molecular weight 39kD and 43kD. The expression of CD94 molecule appears restricted to natural killer (NK) cells and to a T lymphocyte subpopulation including a subset of gamma/+ T-cells, and some CD8+ CD56+ T-cells, mainly V gamma 2/V2. CD94 is found to associate with NKG2 to form a heterodimer which is involved in the inhibition of cell mediated cytotoxicity against cells bearing appropriate MHC class I allotypes. On NK cells the antigen structure apparently functions as a receptor for HLA-B7 on target cells. This interaction is inhibiting cytolytic activity of the NK cells and protects the cells carrying HLA-By. Initially selected for its capacity to inhibit IL2-dependent proliferation of NK-cells. Blocking the receptor on resting NK cells, triggers the Fc-dependent redirected lysis by these cells of HLA-B7 carrying target cells. Fusion Partners: Spleen cells from immunized BALB/c mice were fused with cells of the SP2/0 myeloma cell line. Applications: . Flow Cytometry: Use 10ul of the suggested working dilution to label 1 x 10e6 cells in 100ul. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
