- CD45 or Leukocyte Common Antigen (LCA/T200) family is comprised of at least four isoforms (CD45RA, CD45RB, CD45RC, and CD45RO) of membrane glycoproteins with MW of 180kD-220kD. It consists of a large, heavily glycosylated, amino-terminal extracellular domain of 400-550 amino acids, a single transmembrane region and a large carboxy-terminal cytoplasmic domain of 700 amino acids. CD45/LCA is expressed on hematopoietic cell lines but absent on non-hematopoietic cell lines, normal and malignant non-hematopoietic tissues. The intracellular portion of CD45 proteins has phosphatase activity and is involved in regulation of transmembrane signals, functioning as a leukocyte membrane receptor capable of transducing signals via its phosphotyrosine phosphatase domains. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: Neat; 10ul labels 10e6 cells in 100ul Optimal dilutions to be determined by the researcher. Hybridoma: Spleen cells from immunised DA rats were fused with cells of the Y3/Ag1.2.3 rat myeloma cell line. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
