- Human CD11b (alphaM integrin) complexes with CD18 (beta2 integrin) to form the complement receptor type 3 (CR3) heterodimer which binds to ICAM-1, fibrinogen, C3b, and coagulation factor X. CD11b expression is restricted to leukocytes, mainly to myeloid and NK cells. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 10ug/ml Immunohistochemistry: frozen sections Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 ficoll prepared human peripheral white blood cells were washed and pre-incubated for 5 minutes with 20ul of 250ug/ml human IgG. 2. After which they were incubated for 45 minutes on ice with 80ul of C2262-35R at 10ug/ml. 3. Cells were washed twice and incubated with goat anti-mouse IgG/FITC.. 4. Cells were washed three times, fixed and analyzed by FACS. 5. A 35% sub population of the cells stained positive with a mean shift of 1.12 log10 fluorescent units when compared to a Mouse IgG1 negative control (I1904-78) at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
