- Axin-1 is constitutively expressed and acts as a negative regulator of Wnt expression. It has been demonstrated that mouse Wnt, Wnt3a, is secreted by multiple cell lines and can act and as a growth factor for stem cells. Secreted Wnt3a is highly hydrophobic because it is palmitoylated on an essential cysteine C77; the palmitoylated state of Wnt3a is essential for stem cell signaling. Incubation of stem cells with Wnt3a in tissue culture medium resulted in the increased number of mouse hematopoietic stem cells expressing "primitive" phenotype. Applications: Suitable for use in Immunoprecipitation, Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Positive Control: SW-480 cell lysate Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
