- PD-L1 plays a critical role in induction and maintenance of immune tolerance to self. As a ligand for the inhibitory receptor PDCD1/CD279, PD-L1 modulates the activation threshold of T-cells and limits T-cell effector response (1). The PDCD1/CD279-mediated inhibitory pathway is exploited by tumors to attenuate anti-tumor immunity and facilitate tumor survival (2,3). Through a yet unknown activating receptor, it may costimulate T-cell subsets that predominantly produce interleukin-10 (IL10) (4). Applications: Suitable for use in ELISA, Immunocytochemistry, Immunofluorescence, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (paraffin): 5ug/ml Western Blot: 0.25-4ug/ml Immunofluorescence: 2ug/ml Immunocytochemistry: 1ug/ml Optimal dilutions to be determined by the researcher. Positive Control: HeLa Cell Lysate, Daudi Cell Lysate, Human Stomach Carcinoma Tissue Lysate, Human Tonsil Tissue Molecular Weight: ~33kD (predicted), 37kD (observed) Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
