- Various cellular responses are regulated by the second messengers cAMP and cGMP. Phosphodiesterases, such as PDE10A, eliminate cAMP- and cGMP-mediated intracellular signaling by hydrolyzing the cyclic nucleotide to the corresponding nucleoside 5-prime monophosphate. The deduced 779aa protein has a calculated molecular mass of 88.4kD and contains cGMP-binding sequences and a catalytic domain. Northern blot analysis detected a 10-kb PDE10A transcript in several tissues, with highest levels in fetal brain and kidney, in putamen and caudate nucleus of adult brain, and in thyroid and testis. A 4-kb transcript was also detected in thyroid and testis. mRNA dot blot analysis showed a similar PDE10A expression pattern. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
