- By screening a DC subtraction library, de Saint-Vis et al. (1998) isolated a cDNA encoding DCLAMP. Sequence analysis predicted that the 416aa type I integral membrane protein has an N-terminal signal peptide, followed by a 381aa extracellular domain, a hydrophobic transmembrane domain, and a 10aa cytoplasmic domain containing a conserved GY lysosomal targeting motif, characteristic of LAMP family members. The DCLAMP protein contains multiple N- and O-glycosylation sites. Northern blot and RT-PCR analysis revealed expression of a 3.2kb transcript in appendix, thymus, lymph node, lung, and weakly in spleen, as well as in dendritic cell lines; expression was weak or undetectable in other cell lines. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:100-5000 Western Blot: 1:50-400 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 Immunocytochemistry: 1:50-500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
