- MaxLight™550 is a new Yellow-Green photostable dye conjugate comparable to Alexa Fluor™546, 555, DyLight™549 , Cy3™, TRITC and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (550nm); Emission (575nm); Extinction Coefficient 150,000. This antibody cocktail recognizes two melanoma-specific proteins, which include MART-1 and gp100. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Function of gp100 is not known but it is reported to be a useful marker for melanocytes and melanomas. A cocktail of these two markers is sensitive for labeling formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation. Applications: Suitable for use in Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Frozen & Formalin-fixed: 0.5-1.0ug/ml for 30 minutes at RT (Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10mM Citrate Buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes Optimal dilutions to be determined by the researcher. Positive Control: SK-MEL-13 and SK-MEL-19 Melanoma cell lines; Melanomas Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™550 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
