- Cyclin B1(also called CCNB1 and G2/mitotic-specific cyclin B1) is a regulatory protein involved in mitosis. The gene product complexes with p34(cdc2) to form a serine/threonine kinase holoenzyme complex called the maturation-promoting factor (MPF). Two alternative transcripts have been found a constitutively expressed transcript and a cell cycle-regulated transcript that is expressed predominantly during G2/M phase. The different transcripts result from the use of alternate transcription initiation sites. S126 phosphorylation, the site of phosphorylation recognized, is involved in an auto-phosphorylation of the cdk-cyclin complex. Recognition of this phosphorylated site suggests is useful to detect the active form of the complex and may serve as a marker of mitosis. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: ELISA: 1:50,000 Western Blot: 1:100-1:1000 Immunoprecipitation: 1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
