Weitere Details:: Toll-Like Receptor 4 (TLR4) belongs to the IL-1 receptor/Toll-like receptor superfamily. It is expressed in placenta and peripheral blood leukocytes. TLR4 is the pattern recognition receptor for lipopolysaccharide (LPS). Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1x10e6 human peripheral blood monocytes. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: This antibody has been tested for flow cytometry using human peripheral blood monocytes. 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction. 2. After blocking, 10ul of 145634 was added to up to 1 x 10e6 cells and incubated for 30 minutes at RT. 3. Unbound antibody was removed by washing the cells twice in Flow Cytometry Staining Buffer (Catalog # F4568-10). Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer. 4. The cells were resuspended in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with PerCP-labeled mouse IgG2a antibody. This procedure may need to be modified, depending upon the cell type and final utilization. Individual users may need to titrate to determine the optimal reagent amount for their specific use. Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable for 12 months after receipt at 4°C. Freezing PerCP conjugates will result in a substantial loss of activity. PerCP conjugates are sensitive to light.
