- Growth factor active in angiogenesis, vasculogenesis and endothelial cell growth. Induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and induces permeabilization of blood vessels. Binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin. NRP1/Neuropilin-1 binds isoforms VEGF-165 and VEGF-145. Isoform VEGF165B binds to KDR but does not activate downstream signaling pathways, does not activate angiogenesis and inhibits tumor growth. Applications: Suitable for use in ELISA, Western Blot and Neutralization. Other applications not tested. Recommended Dilution: ELISA (Sandwich): 2-4ug/ml (100ul/well) detects at least 100pg/ml of recombinant human VEGF165 when used with biotinylated anti-human VEGF165 (V2110-03) as the detection antibody at a concentration of 0.25-1ug/ml. Western Blot: 0.25-0.5ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human VEGF165 is 0.25-0.5ng/lane under non-reducing conditions. Neutralization: 0.2ug/ml will yield one-half maximal inhibition of the biological activity of human VEGF (10ng/ml). Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
