- The protein is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Sandwich ELISA: 8-10ug/ml detected at least 1000pg/well of recombinant human IL-10 when used with a biotinylated anti-human IL-10 detection antibody. Western Blot: 0.25-0.50ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human IL-10 is 2-4ng/lane, under reducing or non-reducing conditions. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
