- MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. This antibody recognizes a protein of 25-26kD, identified as the bcl-2 alpha oncoprotein. It shows no cross-reaction with Bcl-x or Bax protein. Expression of bcl-2 alpha oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of bcl-2 alpha protein, whereas the normal or hyperplastic germinal centers are negative. Consequently, this antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express bcl-2 protein and the small number in which the neoplastic cells are bcl-2 negative. Applications: Suitable for use in FLISA, Flow Cytometry, Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-1ug/ml Flow Cytometry: 0.5-1ug/10e6 cells Immunofluorescence: 1-2ug/ml Immunoprecipitation: 1-2ug/500ug protein lysate Immunohistochemistry (Frozen & Formalin fixed paraffin embedded): 0.5-1ug/ml/30min @ RT Optimal dilutions to be determined by the researcher. Positive Control: Jurkat, K562, HL-60, or HeLa Cells. Tonsil or follicular lymphomas. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
