- ATF6, a basic-leucine zipper protein, was isolated by yeast interaction assay as a binding partner to SRF (serum response factor). SRF is a transcription factor, which binds to the serum response element (SRE) in the c-fos promoter and regulates expression of the c-fos gene as well as other immediate-early genes and some tissue-specific genes. ATF6 has also been shown to interact with the endoplasmic reticulum (ER) stress response element (ERSE). ATF6 constitutively expressed as a membrane-bound 90kD protein is directly converted to a 50kD protein in ER-stressed cells. When unfolded proteins accumulate in the ER, ATF6 is cleaved to release its cytoplasmic domain, which enters the nucleus. Applications: Suitable for use in FLISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (Formalin fixed paraffin embedded): 3ug/ml Optimal dilutions to be determined by the researcher. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
