- FTO (Fat mass-and obesity-associated gene) is the responsible gene for mouse 'fused toes' mutation. An association between FTO genotype and type 2 diabetes has been confirmed. The presence of the FTO rs9939609 A-allele was found to be positively correlated with other symptoms of the metabolic syndrome, including higher fasting insulin, glucose, triglycerides, and lower HDL-cholesterol. Applications: Suitable for use in ELISA, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilutions: ELISA (direct and indirect): 1:2000-1:5000 Immunoprecipitation: 5ug/ml/sample in a final reaction volume of 500-1000ul Immunohistochemistry (paraffin sections): 5-10ug/ml Western Blot: 1:2000-1:5000 using ECL. Suggested blocking and dilution buffer is PBST containing 0.05% Tween 20 and 5% skim milk. Suggested incubation time is 1 hour at room temperature Optimal dilutions to be determined by researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt.
