- Neutrophils are the first responders to acute inflammation and play a complex role in the tumor microenvironment (often acting as myeloid-derived suppressor cells). When depleting neutrophils in vivo, researchers commonly use the highly specific Anti-Ly6G antibody. However, attempting to validate this depletion by staining for Ly6G or the broader Gr-1 receptor on a flow cytometer often results in severe epitope masking, leading to inaccurate cell counts and false-negative data. ichorbio’s Neutrophil Depletion & Validation Kit entirely bypasses this structural interference. We pair our Low endotoxin in vivo Anti-Mouse Ly6G (Clone 1A8), which specifically clears neutrophils, with a non-competing validation readout: Anti-Mouse CD11b (Clone M1/70). Because Clone 1A8 binds to the Ly6G receptor and Clone M1/70 independently targets the CD11b (Mac-1) antigen on the neutrophil surface, researchers achieve a highly accurate, masking-free quantification of their in vivo depletion efficacy.
