Phospho-Sure™ RTD Neuronal Cell & Soft Tissue Extraction Buffer from Biosensis

We are pleased to introduce you to our newly released Phospho-Sure™ Ready-to-Dilute (RTD) Neuronal extraction buffer - optimized for the extraction of phosphorylated proteins from neuronal and other soft tissues types.

Biosensis' new Phospho-Sure™ Ready-to-Dilute (RTD) Neuronal extraction buffer extracts the phosphoproteins in a native state without the use of harsh detergents or oxidizers, and it is specially formulated to help maintain phosphoproteins and protect them from degradation better than traditional detergent based extraction buffers (see figure 1).

Phospho-Sure™ RTD is designed as both an isolation and extraction buffer and for best results the intact tissue should be bathed in ice-cold Phospho-Sure™ RTD buffer during excision of the tissue. Once the desired tissue piece is isolated in Phospho-Sure™ RTD it may be transferred to a small volume of ice-cold Phospho-Sure™ RTD for disruption.

Figure 1: Shows the decrease in the decay rate in phosphorylated signals from western blots performed on samples held at room temperature for varying times in both Phospho-Sure™ RTD™ ♢ or traditional lysis buffer preparations.

Figure 2: Western blots of CREB-P, SAP/JNK-P, ERK1/2-P and ERK1/2 from inferior colliculus (ICc). Samples of the adult guinea pig central nucleus of the ICc were homogenized in Phospho-Sure™ RTD™ or traditional Tris-HCL triton X-100 lysis buffer and kept on ice for up to 120 min. Aliquots of the homogenates, each containing 30μg protein, were run on SDS-PAGE. Separated proteins were electrophoretically transferred to the blotting membrane, which was incubated with anti-CREB-P antibody. The blot was stripped and treated with anti-SAP/JNK-P, ERK1/2-P or ERK1/2 antibodies.

Preparation of Phospho-Sure™ RTD™ is very simple. Simply add sterile ddH2O directly to the powdered solution in the supplied bottle, swirl briefly to dissolve the chemicals and use. The material contains no preservatives so sterile technique should always be used.

Neural Cell-Type Specific Markers from Aves Labs

  • Aves Labs' catalogue of neural cell marker antibodies can be used alongside mouse or rabbit antibodies in double-labeling studies.
  • All of their neuronal markers are made against sequences that are 100% shared between mouse and human proteins.
  • In some cases, our neural markers contain mixtures of anti-peptide antibodies generated against different regions of the same gene product, improving signals in your experiments, while still assuring target-specificity
  • Examples of the products available include:

Beta Amyloid Peptide (NT) Choline Acetyltransferase Glutamate Receptor (Metabotropic) Type 2 Neu-N
Catalogue #: ABN Catalog #: CAT Catalog #: ER2 Catalog #: NUN

Aves Labs' neural markers are sold in three ways:

(1) As an individual vial

(2) In a NeuralSampler® Kit (one 200 µl vial of Neural Cell Type-Specific Marker antibody plus 50 µl of a secondary anti-chicken IgY antibody* plus 1.0 ml of BlokHen® blocking reagent designed for use with chicken antibodies)

(3) In a NeuralSampler® Kit (three 200 µl vials of any combination of Neural Cell Type-Specific Marker antibodies, plus three vials (50 µl each) of the secondary antibody of your choice, plus 3.0 mls BlokHen® blocking reagent designed for use with chicken antibodies)

* In the NeuralSampler® kits, you have your choice between Aves' secondary anti-chicken IgY antibodies. These affinity-purified secondary antibodies were generated in goats and are coupled with either fluorescein, biotin, alkaline phosphatase, or horseradish peroxidase. Unconjugated goat anti-chicken IgY antibodies are also available. With the 3-Antibody NeuralSampler® Kit, you may choose any combination of secondary antibody, including three of the same one if you want.

Amylo-Glo Staining Kits for localization of amyloid plaques in brain tissue: from Biosensis

Catalog No: TR-300-AG

The Amylo-Glo® RTD™ “Ready to Dilute” Staining reagent is designed to stain amyloid plaques in tissue sections. This novel marker has several advantages over other conventional markers such as Thioflavin S and Congo Red because of its unique chemical and spectral properties. (L. Schmued et al. (2012) J.Neuroscience Methods 209:120–126).

Amylo-Glo® results in a very bright blue UV excitable stain under physiological conditions that does not bleed through when illuminated with other filters. Its brightness makes it ideal for low magnification quantification studies, while its unique excitation/emission profile and mild staining conditions makes it ideal for combination for multiple immunofluorescent labeling studies.

The reagent can be used with fresh, frozen, and formalin-fixed immunohistochemistry or cytochemistry, and it is good for confocal and multiple labeling because of its high fluorescent intensity and high resistance to photo-bleaching.

Amylo-Glo® RTD™ will make quantitative plaque determination better, easier, and faster.
