- Members of the Frizzled family of proteins serve as receptors for the Wnt signaling pathway and are involved in a variety of developmental processes. Frizzled proteins each contain a divergent N-terminal signal peptide, a highly conserved extracellular cysteine-rich domain, a variable-length linker region, a seven-pass transmembrane domain and a variable-length C-terminal tail. Applications: Suitable for use in Western Blot and Direct ELISA. Other applications not tested. Recommended Dilution: Western Blot: Use at 1-2ug/ml with the appropriate secondary reagents to detect mouse Frizzled-1. Using a colorimetric detection system, the detection limit for rmFrizzled-1 is ~5ng/lane and 1ng/lane under non-reducing and reducing conditions, respectively. Direct ELISA: Used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse Frizzled-1. The detection limit for rmFrizzled-1 is ~5ng/well. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
