- Cyr61, also known as IGFBP-10 and CCN1, is a 50kD secreted matrix- and cell-associated protein that regulates the growth and adhesion of vascular endothelial cells, fibroblasts, and monocytes. Cyr61 interacts with cells that express integrins aVB3, aVB5, aMβ2, and a6B1. Cyr61 is cleaved by plasmin within its VWF domain which generates an N-terminal fragment that is not associated with the matrix but retains the ability to induce endothelial cell migration. Cyr61 induces VEGF upregulation, angiogenesis, and tumorigenesis. Between amino acids 176-281, mouse Cyr61 shares 87% and 97% aa sequence identity with human and rat Cyr61, respectively. Applications: Suitable for use in Direct ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Direct ELISA: 0.5-1ug/mL detection limit for rmCyr61 ~1ng/well Western Blot: 1ug/mL Immunohistochemistry: 5-15ug/mL Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
