- CD45, also known as the leukocyte common antigen (LCA), exists as four different isoforms (180, 190, 205, 220kD) that are generated by alternative splicing of three exons. LCA is found on all cells of hematopoietic origin, except erythrocytes and platelets. CD45 expression is required for T cell and B cell antigen receptor-mediated activation.1-4 Applications: • Flow cytometry • Immunohistochemistry (frozen sections) • Immunoprecipitation • Western blotting Recommended Dilution: Flow Cytometry: 10ul/10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. PE/CY5 combination conjugate is used as a third fluorochrome in multicolor flow cytometry applications when only a single 488nm excitation source is available. Antibodies are covalently coupled using heterobifunctional linking reagents, resulting in an optimal conjugation format for flow cytometry.
